Endoplasmic reticulum (ER) stress occurs when the protein-folding capacity of the ER is overwhelmed, leading to the accumulation of unfolded or misfolded proteins within the ER lumen. The ER is a key organelle responsible for the synthesis, folding, and maturation of proteins destined for secretion, membrane insertion, or organelle function. Various physiological and pathological conditions, including nutrient deprivation, oxidative stress, calcium imbalance, viral infection, and genetic mutations, can disrupt protein folding processes and induce ER stress.
To cope with ER stress, cells activate a signaling network known as the unfolded protein response (UPR). The UPR aims to restore ER homeostasis by increasing the capacity for protein folding, enhancing ER-associated degradation (ERAD) to remove misfolded proteins, and reducing the load of newly synthesized proteins entering the ER. The UPR is mediated by three key ER transmembrane proteins: inositol-requiring enzyme 1 (IRE1), activating transcription factor 6 (ATF6), and protein kinase R-like endoplasmic reticulum kinase (PERK).
