May catalyze alpha-1,3 and alpha-1,4 glycosidic linkages involved in the expression of Vim-2, Lewis A, Lewis B, sialyl Lewis X and Lewis X/SSEA-1 antigens. May be involved in blood group Lewis determination; Lewis-positive (Le(+)) individuals have an active enzyme while Lewis-negative (Le(-)) individuals have an inactive enzyme. Also acts on the corresponding 1,4-galactosyl derivative, forming 1,3-L-fucosyl links.

Figure 5 FUT3 knockdown in RL95-2 cells decreases the IL-1β induced spheroids adhesion and surface expression of sLeX. (A and B) RT-qPCR and western blot analysis indicated that FUT3 siRNA transfection significantly decreased the mRNA and protein expression of the target gene at both the basal and IL-1β (20 ng/ml) stimulated condition, n = 3 per group. (C) FUT3 knockdown decreased the basal and IL-1β induced spheroids adhesion to RL95-2 cells monolayer, n = 9 per group. The percentage of adhesion denotes the proportion of attached HTR-8/SVneo spheroids in the total number of initially transferred spheroids into each chamber. Raw data from the spheroids adhesion experiment can be found in Supplemental Table 2. (D and E) Flow cytometry suggested a substantial reduction of surface sLeX but not LeY expression by FUT3 siRNA transfection, n = 3 per group. Numbers by the representative histograms of isotype control IgM and specific sLeX/LeY staining indicate the positive cells (%). Data represent mean ± SEM.
