二抗
第二抗体是能和抗体结合,即抗体的抗体,简称二抗。其主要作用是检测抗体的存在,放大一抗的信号。二抗是利用抗体是大分子的蛋白质具有抗原性的性质,去免疫异种动物,由异种动物的免疫系统产生的针对于此抗体的免疫球蛋白。二抗一般都带有可以被检测出的标记(如带荧光、放射性、化学发光或显色基团),作用是检测一抗。 如果一抗自己带有可以被检测出的标记时可以不需要二抗,但这样成本很高,因为一种一抗只识别一种底物。所以常见的设计是二抗带上可检测标记,用于检测一抗。当一抗和底物结合后,一个一抗上可同时被多个二抗结合,再加上二抗本身可以设计连接多个显色基团或酶,因此通过二抗可以很容易将检测信号放大,增强检测的灵敏度。
