The NF-kB (nuclear factor kappa-light-chain-enhancer of activated B cells) pathway is a central signaling pathway involved in regulating immune responses, inflammation, cell survival, and proliferation. In its inactive state, NF-kB dimers are sequestered in the cytoplasm by inhibitory proteins known as IkBs (inhibitor of kB). Upon activation by various stimuli, such as pro-inflammatory cytokines, microbial products, or cellular stress, the IkBs are phosphorylated and degraded, allowing NF-kB dimers to translocate into the nucleus. Once in the nucleus, NF-kB regulates the expression of target genes involved in immune and inflammatory responses, cell proliferation, and apoptosis. Dysregulation of the NF-kB pathway has been implicated in various diseases, including inflammatory disorders, autoimmune diseases, cancer, and neurodegenerative diseases.
